Method Description
Salmonella spp. Suitability Testing (qPCR) confirms that the Method can effectively detect Salmonella in a specific sample matrix. The assessment includes analysis of a fortified sample (matrix spike) and a laboratory control sample (LCS). Recovery from the sample is compared to the LCS to evaluate matrix inhibition and ensure reliable detection.

Product-specific suitability per USP <2022> (suitability of the test method in the presence of the product). Three preparations are enriched and analysed side by side: the Product Sample as received, a Matrix Spike (the product inoculated with a low, known level of the target organism, not more than 100 CFU) and a Laboratory Control Sample (the same inoculum without product). Each is reported Presumptive Positive / ND per 25 g; the acceptance criterion is that the Matrix Spike is detected. If the spike is not detected the product inhibits the method, so neutralisers or further dilution are applied and the suitability is repeated.
